Your First Safehouse
The first mission of the game starts after a the introduction scenes in which we see some of Claude's (the protagonist's) backstory: How he came to be in the prison transport and how he was broken out and how Callahan Bridge came to be destroyed.
When finally given control of the situation, you're in your prisoner overalls and the first thing you're told to do is drive your fellow escapee, 8-Ball, to your very first safehouse in the Red Light district. Just follow the pink marker on your mini-map.
Once there, stop in the blue marker to initiate a short cut-scene where both characters change into some normal clothes. Once this is done, 8-Ball suggests that you meet with Luigi, as he may have some work for you. Follow the pink marker once more to the destination just around the corner and stop in the blue marker.
Another cut-scene starts in which 8-Ball goes in to meet Luigi. Shortly after Luigi comes out of the club and gives you your first proper job: Pick up Misty from the clinic and bring her back to the club.
Follow the green marker on the map to the clinic. Once there, stop near Misty to allow her to get into the car.
After the short drive back, Misty gets our the car and the mission finishes, giving you $1500.
With that initial mission completed, an L icon should appear on the mini-map, indicating another available mission from Luigi.
