Mario and Luigi will come across a Shelltop during their travels in Driftwood Shore, who will make you play the Mole Hunt mini-game in order to progress through the game not only once, but twice. After finishing it the second time, you will have the option to play it again for prizes.
When the mini-game begins, you will see various clouds moving around the mini-game arena. These are moles and you need to hit Mario underneath the ground, then have him pop up in the path of the moles. There are different colored moles, each worth different amount of points, and your goal is to get a certain amount (this is predetermined during the story ones). Periodically, a block will appear that will knock all moles still underground up, giving you all the points.
