Deckard Cain
Also known as Cain the Elder, Cain is the last surviving member of the Hoadrim, an order of mages whose duty it was to trap the Prime Evils and keep guard over them forever. Cain is one of the first characters seen in Diablo III, along with his niece LEah as they are investigating the Tristam Cathedral.

Leah
Leah is the adopted niece of Deckard Cain and the daugher of the witch Adria and Aidan the Dark Wanderer. Leah is controlled by an immense power when enraged. She is eventually able to harness and control this ability.

Tyrael
Tyrael is the Archangel of Justice who assists the hero in his quest and helps defeat Diablo. Tyrael is first encountered early in the game when he is discovered to have fallen along with the star that destroyed the Tristam Cathedral. Tyrael has become a mortal and vowed to stop the Prime Evils from destroying Sanctuary.

The titular character, Diablo is the Lord of terror, the youngest and most powerful of the three Prime Evils who rule the Burning Hells.

King Leoric
King Leoric was a lord who declared himself king in the name of the religion Zakarum. He settled in the town of Tristam and unknowingly became a vessel for Diablo. Killed by the Captain of his own army, Diablo resurrected him as the Skeleton King to command the skeletons of the underground.

Izual
Izual is a fallen archangel, appearing in the previous two games in the Diablo franchise who appears as a miniboss in Diablo III.
