You'll get this mission immediately after completing "Wildlife Preserveration". Talk to Roland and he'll send you to Thousand Cuts to talk to the Slab King. He'll give you a note and mention that while the Slab King is a rather brutal man, he owes Roland a favor. Make your way out to Thousand Cuts and be prepared for a fight. Make sure to grab "Poetic License" from Scooter before leaving Sanctuary.

Thousand Cuts
You'll almost immediately wander into a bandit camp when you arrive in Thousand Cuts. You're going to encounter all manner of bandits in this area including Caustic Goliaths, Marauders, Midgets, Psychos, and Buzzards of all kinds. And yes, you do have to kill these guys even though you want to be their ally. There is a waypoint marked on the map that you need to make your way over to. I recommend taking it kind of slow because there's a surprising number of enemies in this camp, espeially as you get closer to your destination.
Make your way across the bridge and towards the large warehouse like structure. Not far from here is a quick mission called "Shoot This Guy In The Face". It's pretty quick and you just have to shoot that guy in the face from close range. Do that and then go up to the front door of the warehouse. You won't be able to get in this way but if you look to the left, you'll see stairs leading up. Go up to the scaffolding and you'll find that you can drop into the warehouse from here.
Initiation
There is not a lot that can be said about this except that it's going to be one long fight. You'll start off with just your standard bandit fair, particularly marauders and work your way up. The space under the Slab King's throne is probably the safest place because none of the enemies from above can hit you here and there's a lot of cover to work with.
After a few waves of Marauders, you'll get some Marauder Badasses. Then you'll start getting rounds with almost nothing but Psycho Midgets. After that, Goliaths will start dropping down almost every round with a group of midgets. When it seems that you are out of enemies, sweep through the bottom area one more time. When you're sure that you are alone, start shooting the guys around the Slab King. There are two Goliaths up there and possibly a couple of Midgets as well. Start by killing the midgets, then enrage one Goliath at a time. When you've killed all of those enemies, the Slab King will come down and introduce himself.
Beacon Defense!
In this section, you need to follow Brick through his camp, destroying the three Beacons that have been scattered around. These beacons are the source of the mortars that are raining down on you. As you go, make sure you watch out for the red circles that are appearing everywhere. When you are moving, make sure you are spring. When you aren't moving, make sure you are right next to one of the beacons or are inside of a shipping crate (or similar).
As you run around with Brick, you're going to encounter your standard Loader enemies. There will be Surveyors and Loaders all over but they shouldn't be too much of a problem. Brick will run up to them and destroy most of them in just a few hits. If you are having trouble, hang back and let Brick handle it. Just don't run too far ahead becuase he'll think that you've fallen behind. There's also a new type of Loader here called the ION Loader which can cause some serious problems. The can put up a dome shaped shield that you can't shoot through but that other Loaders can, putting you at a severe disadvantage. And don't think you're just going to march through that shield because if you do, the ION loader will sap your shields almost instantaneously, making you an easy target. Make sure you prioritize them.
When you get to a beacon, Brick will break the shields down and then you need to destroy it. You can do this just by hitting it with melee attacks. Just repeat this process for each of the three beacons. When they've all been destroyed, Brick will lead you back to the entrance of the camp and tell you that the Slab may still attack you from time to time (because they're stupid) but just kill them, it's ok. Head back to Sanctuary and talk to Roland to finish the mission.
