This mission is available as soon as you arrive in Sanctuary in "The Road to Sanctuary" and is the first in Marcus' elemental weapons quest line.

Find Marcus' shop in town and talk to him to start this mission. He'll lead you down into his shooting range where you will be given a quest weapon that you'll need to equip before shooting the dummy target in the shooting range.
